- Stimulates megakaryocyte platelet production → ↑ platelets in immune thrombocytopenia and aplastic anemia
- Platelet overshoot → thrombosis
- Stimulates the MPL receptor on megakaryocytes
- Raises platelet counts in ITP and liver-disease thrombocytopenia
- Thrombosis risk as counts normalize

- Avatrombopag — Oral TPO-receptor agonist for thrombocytopenia in chronic liver disease or chronic ITP.
- Eltrombopag — Oral thrombopoietin-receptor agonist; cation chelation complicates dosing, while liver injury and thrombosis matter.
