5-HT1F agonist
5-HT1F serotonin receptor agonist
5-HT1F agonist — mechanism card
- On trigeminal neurons → ↓ nociceptive signaling and CGRP (calcitonin gene-related peptide) release
- Aborts migraine without vasoconstriction unless paired with 5-HT1B/1D agonism
- If BBB-crossing: dizziness, somnolence
- Lasmiditan (Reyvow) is a selective 5-HT1F agonist – antimigraine effect similar to 5-HT1B/1D agonist without vasoconstriction
Drugs with this mechanism (3)
- Lasmiditan — A "ditan" 5-HT1F agonist for migraine; no vasoconstriction, causes driving impairment.
- Rizatriptan — A fast-acting triptan for migraine.
- Serotonin — The endogenous ligand at fourteen receptor subtypes; the home of serotonicity — synaptic serotonin elevation.
