Gamma-Glutamyl carboxylase cofactor
Gamma-Glutamyl carboxylase cofactor — mechanism card
- Carboxylates clotting factors II, VII, IX, X to make them functional
- Recycled by VKORC1 — the step vitamin K antagonists block
- High doses bypass VKORC1 blockade → reverses vitamin K antagonist anticoagulation
- Required cofactor for gamma-Glutamyl carboxylase
Drugs with this mechanism (2)
- Menaquinone — Vitamin K2; gamma-carboxylation cofactor.
- Phytonadione — Vitamin K1; clotting-factor carboxylation cofactor and the warfarin antidote.
